What causes fibrous dysplasia?

The exact cause of fibrous dysplasia is not known, but it is believed to be due to a chemical abnormality in the protein of the bone. It may be an inherited disorder. Recent studies indicate that fibrous dysplasia may be caused by a chemical abnormality in a protein in the bone that leads to an overgrowth of bone cells that produce fibrous tissue. The chemical abnormality occurs because of a mutation or change in the structure of the gene that produces the protein. Fibrous dysplasia is thought to be a congenital disorder, meaning that individuals who have it probably had it when they were born. There is no evidence, however, that the disorder can be inherited. In fibrous dysplasia, the medullary bone is replaced by fibrous tissue, which appears radiolucent on radiographs, with the classically described ground-glass appearance. Trabeculae of woven bone contain fluid-filled cysts that are embedded largely in collagenous fibrous matrix, which contributes to the generalized hazy appearance of the bone.
